Detailed description

Plagl1{–8} mice carry a CRISPR/Cas9-generated mutation (8 bp deletion) of a 3' UTR site in the mouse Plagl1 (pleiomorphic adenoma gene-like 1) gene that is required for target-directed degradation of miR-322-5p.

The Plagl1{-22+7} (Stock No. 041171) and Plagl1{-8} (Stock No. 041172) mutations are predicted to cause the same molecular disruption, and experimental results are consistent between the alleles. The donating laboratory treats the two alleles as interchangeable.

Mice with a paternally inherited Plagl1 mutant allele produce elevated levels of miR-322-5p in the embryonic day 18.5 (E18.5) heart and stomach due to impaired target-directed microRNA degradation (TDMD) and are 5.5% smaller than wildtype littermates at E18.5. This body size phenotype persists into adulthood; 8-week-old mice are approximately 7-8% smaller than wildtype littermates. No embryonic growth phenotype is observed when Plagl1 mutant alleles are inherited maternally.

Development

CRISPR-Cas9 methodology was used to create an 8 bp deletion in a 3' UTR site of the mouse Plagl1 gene on mouse Chromosome 10 (ENSMUST00000121646.8). The mutation was created in C57BL/6J embryos. Resultant mice were maintained on a C57BL/6J genetic background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
