Cells from the 8F10 reactive type B T cell line, derived from B:9-23 insulin peptide-immunized NOD mice, have been shown to exhibit strong reactivity for antigen presenting cells (APCs) pulsed with the B:9-23 insulin peptide, while remaining completely unreactive to APCs pulsed with the complete insulin protein. The T cells specifically recognize the type B register 12-20 and completely lack a response to the type A register 13-21. These 8F10 T cell receptor (TCR) transgenic mice were generated using the rearranged TCR &alpha; chain (V&alpha;13.3, TRAV5D-4/TRAJ53) and &beta; chain (V&beta;8.2, TRBV13-2/TRBD2/TRBJ2-7) cloned from 8F10 T cells. Tcra is expressed from the human CD2 promoter and Tcrb is expressed from the mouse Cd4 promoter. The T cell repertoire of these mice is highly skewed to react to the insulin B chain 12-20 epitope. Animals manifest early insulitis and a lack of diabetes onset. The development of CD8+ T cells is impaired in these 8F10 mice, as seen by their reduced number in thymus and spleen.
