Camk2a-tTS mice express a tetracycline-regulated suppressor gene (tTS) under the control of the mouse calcium/calmodulin-dependent protein kinase II alpha (Camk2a) promoter. Camk2a (calcium/calmodulin-dependent protein kinase II alpha) encodes one of four chains of the calcium calmodulin-dependent protein kinase, a major postsynaptic density protein. CAMK2 is required for hippocampal long-term potentiation (LTP) and spatial learning and is able to autophosphorylate resulting in calmodulin-independent activity. tTS encodes a chimeric molecule containing the tetracycline repressor and a transcriptional repressing domain, also known as tetR, which binds tetO, blocking transcription. In the presence of doxycycline, the blocking effect is completely reversed. Hemizygous mice are viable and fertile. When hemizygotes are mated to a second strain carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (TRE; tetO), the target gene expression is repressed and expression can only be initiated by administration of the tetracycline analog, doxycycline (dox). These mice allow repression and inducible expression of genes in forebrain neurons, and may be useful in studying brain disorders such as Alzheimer's disease, Parkinson's disease, and other neurodegenerative diseases.
