Contactin-2 (coded by Cntn2), also referred to as TAG-1, is a transmembrane protein and cell adhesion molecule involved in axon guidance, synapse formation, and myelination. Mutations in this gene are associated with epilepsy and neurodevelopmental impairment. These Tag1EGFP knock-in mice have a loxP-flanked EGFP and stop sequence, followed by an attenuated form of the DTA gene, replacing exon 2 of the Cntn2 gene, while retaining endogenous Cntn2 expression. Heterozygous and homozygous mice are viable and fertile with no overt phenotype. Prior to Cre-recombination, EGFP is expressed in the developing cortex, thalamus, internal capsule and brainstem and colocalizes with endogenous TAG-1. The donating laboratory indicates that EGFP is detectable by immunolabelling. When Tag1EGFP mice are crossed with a strain expressing Cre recombinase, deletion of the loxP-flanked EGFP sequence will result in the expression of diphtheria toxin in the Cre- and TAG-1-expressing cells of the offspring, resulting in ablation of TAS-1 expressing cells. Breeding these Tag1EGFP mice to an Emx1-Cre mouse line resulted in complete loss of all Cre- and TAG-1 expressing cells in the developing neocortex. These mice may be useful for visualization and tracking of TAG-1-positive cells during neuronal development and for studying the role of TAG-1 in axonal guidance, migration, and circuit assembly.
