Description

Cav1^flox mice carry two loxP sites flanking exon 3 of the Cav1 gene. These mice may be useful for generating conditional mutants when studying the role of CAV1 in vesicular and cholesterol trafficking, signal transduction, neurodegeneration and tumorigenesis.

Detailed description

Caveolin 1 is a structural protein for caveolae, and is implicated in regulating nutrient scavenging, lipid accumulation, lipid and glucose metabolism, signal transduction, and mitochondrial function. Cav1^flox mice possess loxP sites flanking exon 3 of the caveolin 1, caveolae protein (Cav1) gene. Homozygous mice are viable and fertile with no overt phenotype. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues, leading to loss of of CAV-1 protein expression. These Cav1^flox may be useful when studying the role of CAV-1 in cellular proliferation, lipid metabolism, cellular tracking, and signal transduction.

Development

The Cav1^flox targeting vector was designed to insert loxP sites flanking exon 3 of the caveolin 1, caveolae protein (Cav1) gene. The targeting construct also included an FRT-flanked neomycin cassette. The construct was electroporated into 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺-derived W9.5 embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The FRT-flanked neomycin cassette was removed by transient expression of FLP recombinase. Chimeric mice were bred to C57BL/6J mice for one generation to confirm germline transmission. The resulting Cav1^flox mice were then maintained on a C57BL/6J background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
