Description

REX3 transgenic mice co-expresses a red fluorescent protein (RFP) driven by the mouse C-X-C motif chemokine ligand 9 (Cxcl9) promotor and a blue fluorescent protein (BFP) driven my the mouse C-X-C motif chemokine ligand 10 (Cxcl10) promoter. These mice may be useful for tracking chemokine expression in immune and inflammatory response pathways.

Detailed description

CXCL9 (also known as MIG, monokine induced by gamma-interferon) and CXCL10 (IP-10, interferon-induced protein of 10kDa) are ligands for the C-X-C motif chemokine receptor 3 (CXCR3). CXCR3 is highly expressed on T cells and natural killer cells and plays an important role in immune cell trafficking, immune responses and inflammatory processes. The REX3 (Reporting the Expression of CXCR3 ligands) transgenic mouse line co-expresses a red fluorescent protein (RFP) driven by the mouse C-X-C motif chemokine ligand 9 (Cxcl9) promotor and a blue fluorescent protein (BFP) driven by the mouse C-X-C motif chemokine ligand 10 (Cxcl10) promoter.

Hemizygous and homozygous mice are viable and fertile. RFP and BFP expression is detectable by flow cytometry. These mice may be useful for tracking the cellular source of CXCL9 and CXCL10 chemokine expression in immune response and inflammatory pathways.

Of note, the transgene integration site and copy number have not been confirmed by the donating laboratory.

Development

The REX3 transgene (Reporting the Expression of CXCR3 ligands) was generated using a BAC clone (RP-24-164O11 BAC). The BAC clone was altered by inserting a red fluorescent protein (RFP) sequence (pTagRFP-N vector, Evrogen), SV40 polyA and an FRT-flanked hygromycin cassette into the ATG start site of the C-X-C motif chemokine ligand 9 (Cxcl9) gene, as well as blue fluorescent protein (BFP) cDNA (pTagBFP-N vector, Evrogen), SV40 polyA and a loxP-flanked kanamycin cassette into the ATG start site of the C-X-C motif chemokine ligand 10 (Cxcl10) gene. Flp and cre expression cassettes (Gene Bridges GmbH) were sequentially electroporated into the BAC construct-containing bacteria to remove the antibiotic resistance genes. The 162kb REX3 construct was linearized and microinjected into the pronucleus of C57BL/6J zygotes. Founder line 1 was established and then subsequently maintained on a C57BL/6J background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J (Stock No. 000664) oocytes.
