Description

pi2^em1Pdz (pi2^-) mice carry partial deletions of the Pirnac2 (piRNA cluster 2) and Gm13817 (predicted gene 13817) loci. This knock-out strain has been used to characterize the contribution of the two non-coding RNAs to mouse fertility and determine the targets of piRNAs produced from Pirnac2.

Detailed description

Argonaute proteins fight viruses, plasmids and transposons, regulate gene expression, or aid DNA replication. Animals produce two distinct types of Argonaute protein: AGO and PIWI. PIWI-clade Argonaute proteins use piRNAs (which are 18-35 nucleotides long) as guides to repress transposons or other coding and noncoding mRNAs. Pirnac noncoding transcripts are expressed post-meiotically in male germ cells, and play a role in regulation of the spermatogenic transcriptome.

pi2^em1Pdz (pi2^-) mice carry a 2,744 bp deletion that includes the promoters of Pirnac2 (piRNA cluster 2) and Gm13817 (predicted gene 13817), the transcriptional start site and part of the first exon of Pirnac2, and the first exon and part of the first intron of Gm13817 on mouse Chromosome 2. This knock-out strain has been used to characterize the contribution of the two non-coding RNAs to mouse fertility and determine the targets of piRNAs produced from Pirnac2.

Homozygotes are healthy and fertile. Adult homozygous males have less progressively motile sperm, with no impact on fertility.

Development

Two sgRNAs (GCT TGA TCG TCA GGG ACT AA and TCA GAG GCT AAG TCC CAT TA) targeting the mouse Pirnac2 piRNA cluster/Gm13817 long non-coding transcript were injected, with Cas9 mRNA, into C57BL/6J embryos. The resulting transgenic mouse line carries a 2,744 bp deletion (chr2:92539403-92542146; mm10) including: the promoter of both genes, and the transcriptional start site and part of the first exon of Pirnac2, and the first exon and part of the first intron of Gm13817. The donating investigator reports the resultant mice were backcrossed to C57BL/6J for 4 generations prior to sending frozen sperm to The Jackson Laboratory. Upon arrival, that frozen sperm is used to fertilize C57BL/6J oocytes (Stock No. 000664) to establish our live colony. Thereafter, males are used to cryopreserve this strain.
