Atoh1-tTA mice express a tetracycline-regulated transactivator gene (tTA2) under the control of the mouse atonal bHLH transcription factor 1 (Atoh1) promoter. Atoh1 encodes a transcription factor that plays a role in the differentiation of subsets of neural cells, such as cochlear hair cells (HCs), the dorsal spinal cord, brainstem, cerebellum, and intestinal secretory cells. When these transgenic mice are bred with mice carrying a gene of interest under the regulatory control of a tetracycline-responsive promoter element (TRE or tetO), the target gene is expressed in granule cell precursors (GCPs) in the developing cerebellum, which can be inhibited with administration of the tetracycline analog, doxycycline (dox). Homozygous mice are viable and fertile with no reported phenotype abnormalities.
