Atoh1-FlpoER mice express a tamoxifen-inducible optimized flpo/ERT2 fusion gene under the control of the mouse atonal bHLH transcription factor 1 (Atoh1) promoter. Atoh1 encodes a transcription factor that plays a role in the differentiation of subsets of neural cells, such as cochlear hair cells (HCs), the dorsal spinal cord, brainstem, cerebellum, and intestinal secretory cells. In this strain, ATOH1 directs expression of the tamoxifen-inducible flpo recombinase specifically to granule cell precursors (GCPs) in the external granule cell layer (EGL) of the developing cerebellum. When transgenic mice are bred with mice containing FRT-flanked sequences, tamoxifen-inducible flpo-mediated recombination will result in deletion of FRT-flanked sequences in the offspring. Homozygous mice are viable and fertile with no reported phenotype abnormalities. When the Atoh1-FlpoER transgene and the R26MASTR allele (Stock No. <a href="https://www.jax.org/strain/018903">018903</a>), which expresses an eGFPcre fusion protein upon flp-induced deletion of a STOP cassette, are combined with a floxed gene such as Gli2 (Stock No. <a href="https://www.jax.org/strain/007926">007926</a>), recombination of the loxP sites occurs in &gt; 98% of GFP+ cells within 3 days of administrating tamoxifen (Tm) at P2. The mutant cells and their progeny can subsequently be identified by the continuous expression of eGFP from the R26 allele. The Atoh1-FlpoER transgene is useful for mosaic mutant analysis of GCPs in the cerebellum and for modeling sporadic cancer after tamoxifen is administered. The donating investigator reports no recombination in the absence of tamoxifen.
