Description

Rorc^tdtomato BAC transgenic mice express a tdTomato fluorescent reporter from the mouse Rorc (RAR-related orphan receptor gamma; RORγt) promoter in conjunction with a Ptprc^a (CD45.1) marker. These mice faithfully mark cells expressing RORγt including all developing thymocytes, Th17 cells, gamma delta (γδ) T cells, and Type 3 innate lymphoid cells (ILC3s).

Detailed description

Rorc^tdtomato BAC transgenic mice express a tdTomato fluorescent reporter from the mouse Rorc (RAR-related orphan receptor gamma; RORγt) promoter. These mice faithfully mark cells expressing RORγt including all developing thymocytes, Th17 cells, gamma delta (γδ) T cells, and Type 3 innate lymphoid cells (ILC3s). This reporter is said to be much brighter than other GFP reporters and since it is not a knock-in, does not disrupt the expression of endogenous Rorc. The mice carry the Ptprc^a (CD45.1 allele) and may be segregating for Pepc^b.

Development

A tdTomato-BGHpA-FRT-neomycin-FRT cassette was introduced via homologous recombination to the open reading frame of the RORγt isoform, 3’ of the ATG start site in BAC clone RP23-263K17. Pronuclear injections of the BAC vector were done in C57BL/6 fertilized zygotes. The neomycin cassette was removed by FLP-mediated recombination leaving one FRT site. Resultant mice were bred with B6.SJL-Ptprc^a Pepc^b/BoyJ (Stock No. 002014) for 8 generations by the donating laboratory. The mice carry the Ptprc^a (CD45.1 allele) and may be segregating for Pepc^b. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize B6.SJL-Ptprc^a Pepc^b/BoyJ oocytes (Stock No. 002014).

Molecular note: Ptprc^a is found in strains SJL/J, STS/A, and DA. Ptprc^b is found in strains C57BL/6, C3H/An, DBA/2, AKR, and many others (J:13367, J:12054, J:12077, J:8603). Twelve nucleotide differences between the a and b alleles have been identified. These base substitutions correspond to five amino-acid changes within the extracellular domain of the encoded protein. These amino-acid differences are clustered in a region that also contains the greatest divergence between mouse and rat sequences (J:22485).

Note that the allele designations originally described were reversed in 1987 (J:8603); all publications prior to 1987 show SJL/J, STS/A, and DA as having the b allele and the C57BL/6J group as having the a allele (J:22341).
