Description

miR-92^fl mice carry two loxP sites flanking the microRNA-92-1(Mir92-1) sequence on chromosome 14. These mice may be useful for generating conditional mutants when studying the regulatory role of microRNA-92-1 in cellular proliferation, differentiation and survival.

Detailed description

MicroRNA 92-1 (coded by Mir92-1) is a short, non-coding RNA that is part of the miR-17-92 cluster and implicated in cell proliferation, apoptosis, and promoting tumor growth in several types of cancer. MicroRNA 92-1 has also been linked to regulating heart, lung and skeletal development. miR-92^fl mice possess loxP sites flanking the Mir92-1 sequence. Homozygous mice are viable and fertile with no overt phenotype. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the Mir92-1 sequence deleted in the cre-expressing tissues. These miR-92^fl may be useful when studying the role of MicroRNA 92-1 in cellular proliferation, differentiation and survival.

Development

The miR-92^fl targeting vector was designed to insert two loxP sites flanking the microRNA 92-1 (Mir92-1) sequence on chromosome 14. The targeting construct also included an FRT-flanked neomycin cassette. The construct was electroporated into 129Sv/Pas-derived embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. Chimeric mice were bred to C57BL/6J mice for one generation to confirm germline transmission. The resulting miR-92^fl mice were then crossed to a FLP-recombinase mouse line to remove the neomycin cassette. miR-92^fl mouse line, with neomycin cassette removed, were backcrossed to C57BL/6J background for 8 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
