Detailed description

T1/ST2 KO mice harbor a mutation generated by replacing exons4-5 of the interleukin 1 receptor-like 1 (Il1rl1) gene with a neomycin cassette, resulting in a null allele. Il1rl1 codes for T1/ST2 which is expressed on the surface of T helper type 2 (Th2) cells and mast cells. The T1/ST2 receptor and its ligand, interleukin 33 (IL-33), are involved in the development of fibrosis, tissue damage, inflammation, and remodeling.

Homozygous T1/ST2 KO are viable and fertile, though it is recommended that these mice be housed in specific pathogen free (SPF) conditions. The T1/ST2 KO mice do not appear to have compromised baseline immune function; however, when exposed to antigens such as S. mansoni, these mice failed to develop significant pulmonary granuloma formation, nor did they demonstrate eosinophil infiltration in the lungs. These mice may be useful when studying inflammatory and autoimmune diseases, cancer, and cardiovascular diseases.

Development

The T1/ST2 knock-out (KO) allele was generated by insertion of a neomycin cassette which replaces exons 4 and 5 of the interleukin 1 receptor-like 1 (Il1rl1). The construct was electroporated into 129P2/OlaHsd-derived E14.1 embryonic stem (ES) cells. Targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric mice were bred with C57BL/6 mice to confirm germline transmission. Heterozygous T1/ST2 KO mice were intercrossed to generate homozygous mice and maintained on a mixed B6;129 background by Dr. Andrew McKenzie.

Some of the T1/ST2 KO mice from the McKenzie laboratory were transferred to Dr. Palmer-Lourenco in 2008. The Palmer-Lourenco laboratory crossed these T1/ST2 KO mice to ROSA-stop-flox-IL33 knock-in (KI) mice. Male germplasm that was homozygous for the T1/ST2 KO allele and homozygous for the ROSA-stop-flox-IL33 KI allele were shipped to The Jackson Laboratory. Upon arrival, the male germplasm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304). The recovered progeny were then backcrossed to C57BL/6NJ for at least one generation to separate the T1/ST2 KO and ROSA-stop-flox-IL33 KI alleles, resulting in two single mutant mouse lines: T1/ST2 KO offered as Stock No. 039982 and the ROSA-stop-flox-IL33 KI offered as Stock No. 039567.

Molecular note: Portions of exons 4 and 5 were replaced by a neomycin selection cassette inserted at the endogenous locus via homologous recombination. The deleted region encoded a portion of the extracellular domain which is common to both membrane bound and soluble forms of the protein. RT-PCR analysis of activated lymphocytes showed an absence of normal transcript in homozygous mutant mice. Cell surface expression of the encoded protien was undetected by flow cytometric analysis of cultured mast cells.
