IEbeta-mAmetrine knock-in/knock-out mice have an insertion of mAmetrine at the ATG start codon in exon 1 of the of the histocompatibility 2, class II antigen E beta (H2-Eb1) gene. H2-Eb1 encodes part of the MHC class II cell surface protein complex and is involved in adaptive immune responses, antigen processing, and presentation of exogenous peptide antigen via MHC class II. C57BL/6 mice can express MHC class IA proteins, but do not express MHC class II IE due to a nonfunctional IE-alpha gene, but the IE-beta gene is fully functional. In these IEbeta-mAmetrine knock-in/knock-out mice, insertion of the mAmetrine sequence abolishes endogenous IE-beta expression. IEbeta-mAmetrine mice are maintained on an albino genetic background to aid in intravital imaging of skin. mAmetrine fluorescence is detectable in MHC class II expressing cells in lymph nodes, spleen, thymus, and lung by direct microscopy, flow cytometry, and two photon intravital microscopy. Homozygous mice are viable and fertile.
