Description

Exon 2 of the solute carrier family 7 (cationic amino acid transporter, y+ system), member 11 (Slc7a11) gene is flanked by loxP sites in this conditional mutant strain. These mice may be useful in studying the functions of Slc7a11 in oxidative stress, cancer biology, neurodegenerative disease, and the blood-brain barrier.

Detailed description

This strain was generated using embryonic stem cells provided by the International Knockout Mouse Consortium (KOMP). Slc7a11 (the solute carrier family 7, cationic amino acid transporter, y+ system) is a member of the Xc- system, which is a sodium-independent amino acid transport system that functions as an antiporter that moves cystine into cells and glutamate out of cells. Exon 2 of the Slc7a11 gene is flanked by loxP sites in this conditional mutant strain. Heterozygous and homozygous mice are healthy, appear normal, and are fertile. When bred to mice that express tissue-specific Cre recombinase, the resulting offspring will have exons 2 deleted in the tissues where Cre is expressed. These SLC7a11 floxed mice may be useful for studying the role of Slc7a11 in oxidative stress, cancer biology, neurodegenerative disease, and the blood-brain barrier.

 

Development

Sperm of STOCK Slc7a11^{tm1a(EUCOMM)Wtsi}/IcsOrl, having the Slc7a11^{tm1a(EUCOMM)Wtsi} targeted allele, were obtained from European Mouse Mutant Archive (EMMA), Germany. The targeting construct incorporates an FRT-lacZ-loxP followed by a neomycin resistance gene, SV40 polyA, a second FRT site, and a second loxP site upstream of exon 2 (chr3:50,389,572; Build GRCm39) of the solute carrier family 7 (cationic amino acid transporter, y+ system), member 11 (Slc7a11) sequence on chromosome 3. A third loxP site is inserted downstream in intron 4 (chr3:50,390,342; Build GRCm39). The construct was electroporated into C57BL/6N-derived JM8.N4 embryonic stem (ES) cells. Targeted ES cells were injected into blastocysts to generate founder mice. Dr. Sandra Howett (Syracuse University) obtained cryopreserved sperm from EMMA and rederived tm1a mice onto C57BL/6NJ. The mice were crossed with FLP deleter mice [129S4/SvJaeSor genetic background from The Jackson Laboratory] to excise the FRT-flanked lacZ and neo cassette, leaving exon 2 flanked by loxP sites. Some frozen sperm was provided to The Jackson Laboratory in 2025. To establish our live colony (without the FLP allele), an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304). Subsequently, The Jackson Laboratory cryopreserved our public bankstock for this strain.

In 2025, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the first generation rederived living colony at The Jackson Laboratory Repository. This showed more than 12 markers segregating as B6;129S. These data suggest that the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6;129S genetic background.

Molecular note: The L1L2_Bact_P cassette was inserted at position 50389572 of Chromosome 3 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 50390342. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).
