Description

Pten-YFP knock-in mice constitutively express EYFP-labeled PTEN (phosphatase and tensin homolog) protein. Pten encodes a tumor suppressor that is found in almost all tissues of the body.

Detailed description

Pten-YFP knock-in mice constitutively express a T2A-EYFP reporter introduced immediately upstream of the mouse Pten (phosphatase and tensin homolog) stop codon. EYFP is easily detectable by flow in secondary lymphoid organ T cells, B cells and myeloid cells. The mice demonstrate no overt phenotype. Heterozygotes and homozygotes are viable and fertile.

Development

A furin cleavage linker-T2A-EYFP-FRT-PGK-Neomycin/Kanamycin-bGH pA-FRT cassette was inserted immediately upstream of the stop codon in exon 9 (the last exon) of the mouse Pten gene via homologous recombination in (129S6/SvEvTac x C57BL/6J) F1-derived embryonic stem cells. To excise the FRT-flanked drug selection cassette from the resultant chimeric animals, mice were bred with R26 FLP mice (Stock No. 003946) which had been backcrossed to C57BL/6J for 13 generations. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
