Description

Dusp26 knock-out (KO) mice have a β-gal gene and a neomycin cassette replacing exon 3 of the dual specificity phosphatase 26 (Dusp26) gene, abolishing endogenous gene expression. These mice may be useful for studying the role of DUSP26 in mitochondrial function and oxidative stress response pathways, specifically in neurodegenerative diseases and various cancers.

Detailed description

Dual specificity phosphatase 26 (Dusp26) encodes an atypical phosphatase that mediates dephosphorylation of a range of substrates, including MAP kinases and p53 protein. DUSP26 has been implicated in regulation of mitochondrial respiration and oxidative stress as well as neuronal cell proliferation. These Dusp26 knock-out (KO) mice have a β-gal gene and a floxed neomycin cassette replacing exon 3 of the dual specificity phosphatase 26 (Dusp26) gene, abolishing endogenous gene expression.

Homozygous mice are viable and fertile with no overt phenotype. LacZ expression is detectable throughout the central nervous system and retinal tissue of homozygous Dusp26 KO mice. These mice may be useful for studying the role of DUSP26 in mitochondrial function and oxidative stress response pathways, specifically in neurodegenerative diseases and various cancers.

Of note, the donating investigator indicates that the floxed neomycin cassette remains intact.

Development

The Dusp26 knock-out (KO) targeting vector was designed to insert a β-galactosidase gene (lacZ) immediately downstream of the ATG start codon (exon 3) of the dual specificity phosphatase 26 (Dusp26) gene. The construct also contained a loxP-flanked neomycin cassette following the LacZ sequence. The insertion results in ablation of endogenous Dusp26 expression. This construct was electroporated into 129S2/SvPas-derived D3 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and the resulting chimeric mice were bred to C57BL/6J (Stock No. 000664) mice to confirm germline transmission. These Dusp26 KO mice, with floxed neomycin cassette intact, were further backcrossed to C57BL/6J for 8 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Of note, the donating investigator confirmed that, during backcrossing, the Y chromosome was fixed on the C57BL/6J background.
