Description

R26-Sall4 animals express a flag-tagged Sall4 isoform "a" (Sall4a) and a hemagglutinin-tagged Sall4 isoform "b" (Sall4b) in addition to an FRT-flanked IRES-EGFP reporter from a CAG promoter upon Cre recombinase-mediated excision of a loxP-flanked neomycin cassette. This strain may be useful in studies of embryonic development and cancer.

Detailed description

Spalt like transcription factor 4 (Sall4) encodes a transcription factor and is known to participate in the pluripotency network of embryonic stem cells. It is highly expressed in early-stage post-implantation mouse embryos, then rapidly downregulated during embryonic development. Sall4 is required for limb and posterior axial skeleton development. SALL4 expression is normally absent in most adult tissues, but is activated in various human cancers, including leukemia, germ cell tumors, hepatocellular carcinoma, gastric cancer, colorectal carcinoma, esophageal squamous cell carcinoma, breast cancer, endometrial cancer, lung cancer, and glioma.

R26-Sall4 animals express a flag-tagged Sall4 isoform "a" (Sall4a) and a hemagglutinin-tagged Sall4 isoform "b" (Sall4b) in addition to an FRT-flanked IRES-EGFP reporter from a CAG promoter upon cre recombinase-mediated excision of a loxP-flanked neomycin cassette. The mice demonstrate no overt phenotype. Heterozygotes and homozygotes are both viable and fertile.

Crosses with mesoderm-specific Cre recombinase mice (Tg(T-cre)1Lwd; Harwell) were used to assess whether continued expression of Sall4 interferes with normal embryonic development. In contrast to severe defects caused by the loss of Sall4 expression, overexpression of Sall4 results in normal morphology and pattern in embryos and neonates.

Development

A CAG-loxP-stop (neomycin)-HBB 5'U-loxP-Flag-Sall4a-T2A-HA-Sall4b-FRT-IRES-EGFP-FRT-bGH polyA cassette was introduced to intron 1 of the mouse Gt(ROSA)26Sor gene via homologous recombination in (129X1/SvJ x 129S1/Sv)F1- Kitl⁺-derived R1 embryonic stem (ES) cells. Sall4a and Sall4b represent the two known Sall4 isoforms "a" and "b", respectively. Resultant chimeric animals were bred to C57BL/6J inbred mice for 9 generations by the donating laboratory (see SNP note below). Upon arrival, the colony was bred to C57BL/6J (Stock No. 000664) for at least one generation. Sperm was subsequently cryopreserved.

in 2026, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. One of the 43 markers throughout the genome on Chromosome 6 was segregating with 129, likely a passenger segment from Gt(ROSA) mutation.
