&gamma;&delta; (gamma delta; gd) T cells represent a substantial fraction of intestinal lymphocytes at homeostasis, but they also constitute a major lymphocyte population infiltrating colorectal cancers (CRCs). Vg4KO (V&gamma;4-) mice carry a CRISPR/cas9-generated knock-out of the mouse T cell receptor gamma, variable 4 (Trgv4) gene. The allele incorporates a 34 bp deletion. Alternative TCR rearrangement is not detected in homozygotes. Homozygotes show a significant reduction of Vg4 gd T cells in blood, lung, intestine, and skin. Heterozygous animals show an intermediate phenotype when compared with wildtype littermate controls. Homozyous mice treated with azoxymethane&ndash;dextran sodium sulphate (AOM-DSS) in an induced model of colitis-associated CRC (CAC) do not show differences in tumor number, tumor size, or tumor-infiltrating gd T cells (aside from V&gamma;4) compared with heterozygous littermate controls.
