Description

Afp-GFP transgenic mice are designed to have a fluorescent protein under control of the the alpha fetoprotein (Afp) promoter/enhancer sequences. These mice may be useful for tracking endodermal lineages during embryogenesis.

Detailed description

Afp-GFP transgenic mice are designed to have the alpha fetoprotein (Afp) promoter/enhancer sequences driving expression of a fluorescent protein. Homozygous transgenic mice are viable and fertile with no overt phenotype. visceral endoderm, yolk sac endoderm, a subset of fetal liver hepatocytes, and the epithelium of the gut and pancreas. Fluorescence may be detected by flow cytometry, immunofluorescence, and direct fluorescence. Mice that are homozygous for the transgene are viable and fertile with no overt phenotype. Homozygous mice have a higher transgene expression level than hemizygotes, resulting in brighter fluorescence. These transgenic mice may be useful for visualizing morphogenesis and tissue rearrangements in the developing embryo.

Note on the fluorescent protein in the Afp-EGFP transgene: The Jackson Laboratory genotyping assay Tg(Afp-EGFP)#Maba, utilizing primers in the Afp promoter and just after the expected GFP start codon, identifies transgene carrier (hemizygous) and noncarrier (wildtype) mice. However, transgene carrier mice type as negative via our robust Generic GFP genotyping assay and our SNP Panel GFP. In 2025, advanced quality control analyses by The Jackson Laboratory concluded that the Afp promoter is followed by an atypical variant of fluorescent protein (e.g., an uncommon GFP).

Development

The Afp-GFP transgene are designed to have an enhanced green fluorescent protein (EGFP) driven by the mouse alpha fetoprotein (Afp) promoter/enhancer sequences, followed by an SV40 polyadenylation site (polyA). The ~9.6kb transgene was microinjected into (C57BL/6xC3H)F1 fertilized oocytes, and mice were bred to outbred ICR mice to establish a colony. The transgene integration site and copy number has not been determined by the donating laboratory. Upon arrival at The Jackson Laboratory in 2024, frozen sperm was recovered using C57BL/6J inbred oocytes (Stock No. 000664) to establish the colony.
