Description

ε-globin::H2B-EGFP transgenic mice express an H2B-EGFP fusion protein under the control of the human ε-globin minimal protomer and a truncated human micro-locus control region. These mice may be useful for chromatin-localized fluorescent expression in nuclei of primitive erythroid cells (EryP).

Note that the donating investigator has also made a similar transgenic line available expressing an H2B-CFP (cyan fluorescent protein) fusion protein under the control of the human ε-globin minimal promoter (Stock No. 039609).

Detailed description

ε-globin::H2B-EGFP transgenic mice express an H2B-EGFP fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene) under the control of the human ε-globin minimal protomer and a truncated human micro-locus control region. Hemizygous and homozygous transgenic mice are viable and fertile with no overt phenotype. Fluorescent expression is detectable in the nuclei of primitive erythroid cells (EryP) in the yolk sac (~E6.5-9.0), in the fetal livers between embryonic day E10.5-E14.5 and using flow cytometry, in the circulation throughout the rest of gestation. Fluorescence can be measured by flow cytometry and immunofluorescence. ε-globin::H2B-EGFP transgenic mice are useful for chromatin-localized fluorescent expression in nuclei of primitive erythroid cells (EryP).

Development

The ε-globin::H2B-EGFP transgene was designed with the human embryonic (ε) globin minimal promoter (HBE1) driving expression of an H2B-EGFP fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] with a C-terminal Enhanced Green Fluorescent Protein gene). The construct also contains a human embryonic (ε) globin 3' splice and polyA sequence and a truncated human micro Locus Control Region. The ~4.7kb transgene was microinjected into outbred B6D2F1 fertilized oocytes, and mice from founder line number 55 were mated to outbred ICR mice to establish a colony. The transgene integration site and copy number have not been determined by the donating laboratory. Upon arrival at The Jackson Laboratory in 2024, an aliquot of frozen sperm was recovered using C57BL/6J inbred oocytes (Stock No. 000664) to establish our live colony.
