Connexin 37^LacZ (Cx37^LacZ) mice express a lacZ reporter from the mouse gap junction protein, alpha 4 (Gja4) promoter. Deletion of the majority of exon 2 is predicted to have created a null allele.

Detailed description

Connexin 37^LacZ (Cx37^LacZ) mice express a lacZ reporter from the mouse gap junction protein, alpha 4 (Gja4) promoter. Deletion of the majority of exon 2 is predicted to have created a null allele. No overt phenotype is observed in heterozygous animals. Homozygous animals have not been examined [July 2024], though they are anticipated to phenocopy Gja4 knock-out mice (Stock No. 025698). Additionally, homozygous females will likely be infertile.

Development

The donating laboratory acquired the "knockout-first" Gja4^{tm1a(KOMP)Wtsi} (IMPC Project 23979; KOMP ID 056038) embryonic stem-cell line from the International Mouse Phenotyping Consortium (IMPC). This allele carries an FRT::LacZ::loxP::Neo::FRT::loxP cassette in intron 1 of the mouse Gja4 gene and a loxP site in the 3' UTR within exon 2. It was created in C57BL/6N-A^tm1Brd-derived JM8A1.N3 embryonic stem (ES) cells. Mouse embryos were injected with the ES cells and resultant chimeric male mice were crossed with C57BL/6NCrl females. A single founder was crossed with Actb:Cre (Stock No. 019099) to excise the loxP-flanked sequence, removing the neomycin selection element and the majority of exon 2 (including the entirety of the Gja4 coding sequence). An En2 splice acceptor::IRES:lacZ element immediately downstream of exon 1 was retained, producing an endogenous locus reporter of Gja4 expression.

This strain was backcrossed to C57BL/6NCrl and C57BL/6J inbred mice for a total of 7 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Molecular note: The L1L2_Bact_P cassette was inserted at position 127205644 of Chromosome 4 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 127207233. The critical exon(s) is/are thus flanked by loxP sites. The cre expression without flp expression resulted in a reporter knockout mouse.
