Description

Connexin 37^Flox (Cx37^fl) mice carry a conditional knock-out allele in which the majority of exon 2 of the mouse gap junction protein, alpha 4 (Gja4) gene is flanked by loxP sites.

Detailed description

Connexin 37^Flox (Cx37^fl) mice carry a conditional knock-out allele in which the majority of exon 2 of the mouse gap junction protein, alpha 4 (Gja4) gene is flanked by loxP sites. Cre recombinase-mediated excision of the floxed region results in a knock-out of the entirety of the Gja4 coding sequence.

Homozygous floxed animals are viable and overtly indistinguishable from wildtype mice. Homozygous males are fertile. Homozygous females exhibit impaired fertility reminiscent of the female infertility observed in a Gja4 knock-out strain (Stock No. 025698), but express GJA4 (Connexin 37) protein and do not show any additional knock-out-like characteristics.

Development

The donating laboratory acquired the "knockout-first" Gja4^{tm1a(KOMP)Wtsi} (IMPC Project 23979; KOMP ID 056038) embryonic stem-cell line from the International Mouse Phenotyping Consortium (IMPC). This allele carries an FRT::LacZ::loxP::Neo::FRT::loxP cassette in intron 1 of the mouse Gja4 gene and a loxP site in the 3' UTR within exon 2. It was created in C57BL/6N-A^tm1Brd-derived JM8A1.N3 embryonic stem (ES) cells. Mouse embryos were injected with the ES cells and resultant chimeric male mice were crossed with C57BL/6NCrl females. A single founder was crossed with R26:FLPe (Stock No. 009086) to excise the FRT-flanked sequence, leaving exon 2 flanked by loxP sites. This strain was maintained on a mixed C57BL/6NCrl and C57BL/6J genetic background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Allele

Symbol: Gja4^{tm1c(KOMP)Wtsi}
Name: targeted mutation 1c, Wellcome Trust Sanger Institute
MGI accession ID: MGI:7664576
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Marker symbol: Gja4
Marker name: gap junction protein, alpha 4
Marker synonyms: Cnx37; connexin 37; CX37; CXN37; Cxnh1; Gja-4
Chromosome: 4
Strain of origin: C57BL/6N-A^tm1Brd
Molecular note: The L1L2_Bact_P cassette was inserted at position 127205644 of Chromosome 4 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 127207233. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele.
