Description

p50(f/f) mice have loxP sites flanking exon 4 of the Nfkb1 gene, making them useful for studying the role of NF-κB in regulating the immunological responses and in the development of targeted immunotherapies.

Detailed description

p50(f/f) mice have loxP sites flanking exon 4 of the nuclear factor of kappa light polypeptide gene enhancer in B cells 1, p105 (Nfkb1) gene. Nfkb1 encodes a transcription factor that is activated by various intra- and extra-cellular stimuli related to many biological processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 4 deleted in the cre-expressing tissues.

When bred to LysMcre transgenic mice (Stock No. 004781), the absence of host p50 in in myeloid cells slows tumor growth.

Development

The L1L2_gt0 cassette was inserted at a position on Chromosome 3 upstream of exon 4 of the nuclear factor of kappa light polypeptide gene enhancer in B cells 1, p105 (Nfkb1) gene. The cassette is composed of an FRT-flanked β-galactosidase/neomycin resistance gene fusion (lacZ/neo), and a polyadenylation site (SV40 polyA). This was followed by loxP sites flanking exon 4. The mutation was generated in C57BL/6N derived JM8.F6 embryonic stem (ES) cells, and correctly targeted ES cells were injected into blastocysts. The resulting chimeric mice were bred to ROSA26-FLPo mice (Stock No. 012930) to remove the lacZ/neo cassette and then subsequently mice were bred together to remove the FLPo expressing transgene. These p50(f/f) mice were bred to LysMcre mice (Stock No. 004781) and mice were further maintained on a C57BL/6 genetic background by continuous breeding of p50(f/f);LysMcre with p50(f/f) mice. Cryopreserved sperm was sent to The Jackson Laboratory. To establish our live colony, an aliquot of frozen sperm was used to C57BL/6NJ oocytes (Stock No. 005304). The LysMcre mutant allele was removed from the colony.

Molecular note: The L1L2_gt0 cassette was inserted at position 135368412 of Chromosome 3 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT flanked lacZ/neomycin sequence followed by a loxP site. An additional loxP site is inserted downstream of the targeted exon(s) at position 135369235. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).
