Trpm8^T2a-FlpO knock-in mice express codon-optimized Flp recombinase (FlpO) from the mouse transient receptor potential cation channel, subfamily M, member 8 (Trpm8) promoter enabling the excision of FRT-flanked sequences in Trpm8-expressing cells. These mice have been useful in studies of C-Cold dorsal root ganglion (DRG) somatosensory neurons.

Detailed description

Trpm8^T2a-FlpO knock-in mice express codon-optimized Flp recombinase (FlpO) from the mouse transient receptor potential cation channel, subfamily M, member 8 (Trpm8) promoter enabling the excision of FRT-flanked sequences in Trpm8-expressing cells, including dorsal root ganglion (DRG) neurons in adults, and in some non-neuronal cells during development. It is unknown whether there is expression in the central nervous system. These mice have been useful in studies of C-Cold DRG somatosensory neurons.

Expression of the targeted Trpm8 gene is not disrupted. Homozygotes are viable and fertile and demonstrate no overt phenotype.

Development

A T2a::FlpO::TGA (Stop)::loxP::PGK-Neo::pA::loxP cassette was introduced directly upstream of the stop codon in the last annotated exon of Trpm8 (NC_000067.7) via homologous recombination in B6129F1 hybrid-derived embryonic stem (ES) cells. The neomycin selection cassette was left intact without any apparent effect on phenotype. Resultant mice were backcrossed to CD1 outbred mice and the line was maintained on the mixed genetic background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
