Description

Sstr2^T2a-CreER knock-in mice express tamoxifen-inducible Cre recombinase from the mouse somatostatin receptor 2 (Sstr2) promoter enabling the excision of loxP-flanked sequences in Sstr2-expressing cells. These mice have been useful in studies of a calcitonin gene related peptide (CGRP)α⁺ subtype of dorsal root ganglion (DRG) somatosensory (C-Heat) neurons.

Detailed description

Sstr2^T2a-CreER knock-in mice express tamoxifen-inducible Cre recombinase from the mouse somatostatin receptor 2 (Sstr2) promoter enabling the excision of loxP-flanked sequences in Sstr2-expressing cells, including dorsal root ganglion (DRG) neurons, and potentially in glia of the DRG and spinal cord. These mice have been useful in studies of calcitonin gene related peptide (CGRP)α⁺ subtype of DRG somatosensory (C-Heat) neurons.

Expression of the targeted Sstr2 gene is not disrupted. Homozygotes are viable and fertile and demonstrate no overt phenotype.

Development

A T2a::CreERT2::TGA (Stop)::FRT::PGK-Neo::pA::FRT cassette was introduced directly downstream of the coding exon containing the methionine start codon for Sstr2 (NC_000077.7) via homologous recombination in B6129F1 hybrid-derived embryonic stem (ES) cells. The neomycin selection cassette was left intact without any apparent effect on phenotype. Resultant mice were backcrossed to CD1 outbred mice and the line was maintained on the mixed genetic background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
