Smr2^T2a-Cre knock-in mice express Cre recombinase from the mouse submaxillary gland androgen regulated protein 2 (Smr2) promoter enabling the excision of loxP-flanked sequences in Smr2-expressing cells. These mice have been useful in studies of a calcitonin gene related peptide (CGRP)ζ⁺ subtype of dorsal root ganglion (DRG) somatosensory (Aδ-high-threshold mechanoreceptor (HTMR)/Heat) neurons.

Detailed description

Smr2^T2a-Cre knock-in mice express Cre recombinase from the mouse submaxillary gland androgen regulated protein 2 (Smr2) promoter enabling the excision of loxP-flanked sequences in Smr2-expressing cells, including adult dorsal root ganglion (DRG) neurons and a small number of spinal cord neurons during development. These mice have been useful in studies of a calcitonin gene related peptide (CGRP)ζ⁺ subtype of DRG somatosensory (Aδ-high-threshold mechanoreceptor (HTMR)/Heat) neurons.

Expression of the targeted Smr2 gene is not disrupted. Homozygotes are viable and fertile and demonstrate no overt phenotype.

Development

A T2a::Cre::TGA (Stop)::FRT::PGK-Neo::pA::FRT cassette was introduced directly upstream of the stop codon in the last annotated exon of Smr2 (NC_000071.7) via homologous recombination in B6129F1 hybrid-derived embryonic stem (ES) cells. The neomycin selection cassette was left intact without any apparent effect on phenotype. Resultant mice were backcrossed to CD1 outbred mice and the line was maintained on the mixed genetic background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
