Description

GODZ-floxed mice possess loxP sites flanking exon 3 of the zinc finger, DHHC domain containing 3 (Zdhhc3) gene. These mice may be useful for generating conditional mutants when studying the role of Golgi-specific DHHC (Asp-His-His-Cys) zinc finger protein (GODZ) in cre-expressing tissues, specifically as it relates to posttranslational protein modification, by palmitoylation.

Detailed description

Zdhhc3 (zinc finger, DHHC domain containing 3) encodes an S-acyltransferase, referred to as Golgi-specific DHHC (Asp-His-His-Cys) zinc finger protein (GODZ), that regulates S-palmitoylation. This reversible posttranslational lipid modification of proteins allows for control of protein-membrane associations and trafficking. These GODZ floxed, mice possess loxP sites flanking exon 3 of the Zdhhc3 gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues. These GODZ floxed mice may be useful for studying the role of Zdhhc3 in posttranslational protein modifications, protein function and cell signaling.

Development

C57BL/6N-derived JM8A3.N1 embryonic stem (ES) cells (clone HEPD0531_1_B09 OR HEPD0531_1_B12) bearing the Zdhhc3^{tm1a(EUCOMM)Hmgu} targeted allele were obtained from The European Conditional Mouse Mutagenesis Program (EUCOMM). The targeting cassette is composed of an FRT site followed by lacZ sequence and an initial loxP site upstream of exon 3 of the zinc finger, DHHC domain containing 3 (Zdhhc3) gene. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 3. Targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred with ROSA26-FLPe mice (Stock No. 009086) to excise the FRT site flanked lacZ-loxP-neo cassette. Heterozygous GODZ floxed mice, with neo cassette removed and floxed exon 3 intact, were maintained on a C57BL/6J background by the donating laboratory (see SNP note below). Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).



In 2025, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Five of the 43 markers throughout the genome were segregating with 129. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.

A third loxP site is inserted downstream of the targeted exon(s) at position 122918641. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).
