Dec2 fl/fl mice have loxP sites flanking the promoter and exons 1-3 of the Bhlhe41 gene. These mice may be useful when studying circadian rhythm and the T_H2 cell development and function.

Detailed description

Dec2 fl/fl mice have loxP sites flanking the promoter and exons 1-3 of the basic helix-loop-helix family, member e41 (Bhlhe41) gene. Dec2 encodes a transcriptional repressor with widespread expression that is involved in the control of circadian rhythm and cell differentiation. DEC2 is also selectively expressed in the T helper type 2 (T_H2) subset of T helper cells where it is involved in lineage commitment and differentiation, as well as chronic inflammation associated with tissue repair, allergies, and asthma.

Homozygous Dec2 fl/fl mice are viable and fertile. When bred to CMV-cre mice (Stock No. 006054), Dec2 mRNA in bone marrow of resulting Dec2^-/- mice is not detectable by RT-PCR and DEC2 protein in splenocytes is absent. The development of lymphoid populations in thymus and spleen appears normal in Dec2^-/- mice, although, CD4⁺ T cells exhibit reduced proliferation and IL-2 production compared with wild-type cells.

Development

The Dec2 fl targeting vector was designed to insert a loxP site followed by a frt-flanked neomycin resistance (neo) cassette, and a second loxP site, upstream of the promoter of the basic helix-loop-helix family, member e41 (Bhlhe41) gene. A third loxP site was inserted downstream of exon 3. The construct was electroporated into 129S6/SvEvTac-derived TC-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the chimeric mice were bred to B6N.ROSA26::FLPe mice (Stock No. 016226) to remove the neo cassette. Resulting Dec2 fl offspring were bred to C57BL/6J mice (Stock No. 000664) for at least 6 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
