Description

The NFE2L1 floxed mice possess a loxP-flanked terminal exon of the nuclear factor, erythroid derived 2,-like 1 (Nfe2l1) gene. It is a member of the basic leucine zipper (bZIP) family transcription factor that has a conserved CNC domain first noted in the Drosophila cap and collar (CNC) gene. NFE2L1 is expressed ubiquitously and essential for development. Mouse homozygous for NFE2L1 floxed mutation is normal, viable, and fertile unless crossed with cre-recombinase expressing mice. This line will be useful for studies of the tissue/cell-specific functions of NFE2L1 in adult organs.

Detailed description

The NFE2L1 floxed mice possess a 5' loxP site inserted into the intron 3, and the neomycin cassette sequence followed by a 3' loxP downstream of the terminal exon (exon 4) of the nuclear factor, erythroid derived 2,-like 1 (Nfe2l1) gene. It is a member of the basic leucine zipper (bZIP) family transcription factor with a conserved CNC domain first noted in the Drosophila cap and collar (CNC) gene. NFE2L1 is expressed ubiquitously and is essential for development. The homozygous mice developed normally, were fertile, and showed no obvious abnormalities. The donating laboratory reported that although the IRES-GFP was incorporated in the targeting construct for tracking recombinant cells, GFP expression was too weak to detect in their analysis. When crossed to alb-cre mice (Stock No. 035593) expressing liver-specific cre recombinase, somatic inactivation of Nfe2l1 resulted in steatosis, apoptosis, necrosis, inflammation, and liver fibrosis leading to hepatic neoplasia.

Of note, the donating investigator has donated NFE2L1 KO mice as Stock No. 039469. This line will be useful for the tissue/cell-specific functions of NFE2L1 in adult organs.

Development

The NFE2L1 floxed allele for the locus was generated by a targeting construct containing a 5' loxP site inserted into the Nfe2l1 intron 3, and a phosphoglycerate kinase 1 (PGK-1) promoter-driven neomycin resistance gene (Neo) cassette followed by a 3' loxP site downstream of the terminal exon (exon4). An internal ribosome entry site (IRES)-GFP was also incorporated at the 3' of the neo-cassette. The construct was electroporated into 129X1/SvJ-derived JM-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient mouse blastocysts and the resulting chimeric male mice were bred to C57BL/6J mice. The NFE2L1 floxed line was maintained on a B6;129X1 mixed background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
