Tnc^CreERT2 knock-in (KI) mice express a CreER fusion protein driven by the tenascin C (Tnc) endogenous promoter, while retaining endogenous Tnc expression. When these mice are bred with mice containing a loxP-flanked sequence of interest, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the floxed sequences in the Tnc-expressing cells of the offspring. These mice may be useful when studying cell proliferation and migration as well as epithelial-mesenchymal transition (EMT).

Detailed description

Tenascin C (encoded by Tnc) is an extracellular matrix glycoprotein involved in mediating cell-matrix interaction, cell proliferation and motility. This protein is upregulated in developing and remodeling tissues as well as in various tumors. These Tnc^CreERT2 knock-in (KI) mice express a CreER fusion protein driven by the tenascin C (Tnc) endogenous promoter, while retaining endogenous Tnc expression. Heterozygous mice are viable and fertile. The donating laboratory has not attempted to make the strain homozygous. Restricted to the cytoplasm, Cre-ERT can only gain access to the nuclear compartment after exposure to tamoxifen. When Tnc^CreERT2 KI mice are bred with mice containing loxP-flanked sequences, tamoxifen-inducible Cre-mediated recombination will result in deletion of the floxed sequences in the cre-expressing cells of the offspring.

Development

The Tnc^CreERT2 knock-in (KI) targeting vector was designed to insert a cre/ERT2 fusion protein into the tenascin C (Tnc) locus. Specifically, this vector includes (from 5' to 3') an FRT-flanked Neomycin-STOP, a cre/ERT2 fusion gene (Cre recombinase fused to a human estrogen receptor ligand binding domain) and rabbit beta-globin gene polyadenylation signal. The construct was electroporated into (C57BL/6NTac x 129S6/SvEvTac)F1-derived BA1 embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were identified by genotyping and bred to a C57BL/6 FLP mouse line to remove the neomycin-STOP cassette. The Tnc^CreERT2 mice KI, with neomycin cassette excised, were backcrossed to FVB/N mice for 3 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize FVB/NJ (Stock No. 001800) oocytes.
