Description

Fancj^HA knock-in mice carry an insertion of a 6xHIS-HA epitope tag immediately upstream of the endogenous Brip1 stop codon. These mice may be useful for studying the role of FANCJ in DNA repair.

Detailed description

The targeted gene, BRCA1 interacting protein C-terminal helicase 1 (Brip1), encodes a DNA-dependent ATPase involved in double-strand break repair also referred to as FANCJ. These Fancj^HA knock-in mice carry an insertion of a 6xHIS-HA epitope tag immediately upstream of the endogenous Brip1 stop codon. Homozygous mice are viable and fertile with no overt phenotype. These mice may be useful for studying the role of FANCJ in DNA repair and genome stability.

Development

The Fancj^HA allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. A single guide RNA (5'-TGGGCAGAAGTGAAAGTCGA-3') was used to insert 6xHis-HA epitope tag sequences, and a mutated PAM sequence, immediately upstream of the endogenous stop codon of the BRCA1 interacting protein C-terminal helicase 1 (Brip1) gene on chromosome 11. The sgRNAs, ssDNA (repair template), and the cas9 nuclease mRNA were introduced into the cytoplasm of zygotes derived from C57BL/6J mice with well recognized pronuclei. Correctly targeted pups were identified by PCR and sequencing. The resulting Fancj^HA mice were backcrossed to C57BL/6J background for at least 2 generations to reduced off-target events by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
