Description

Robo3^IRES-CreERT2 knock-in/knock-out mice express tamoxifen-inducible Cre recombinase from the mouse roundabout guidance receptor 3 (Robo3) promoter in Robo3-expressing cells. This strain has been useful in studies of projection neurons of the anterolateral pathway which conveys sensations of pain, temperature, and touch information from the spinal cord to the brain.

Detailed description

Robo3 (roundabout guidance receptor 3) is transiently expressed in developing commissural neurons, including projection neurons (PNs) of the anterolateral pathway which conveys convey sensations of pain, temperature, and touch information from the spinal cord to the brain.

Robo3^IRES-CreERT2 knock-in/knock-out mice express tamoxifen-inducible cre recombinase from the mouse Robo3 promoter in Robo3-expressing cells. Expression in the spinal cord and brain has been examined. Expression of the targeted Robo3 gene is knocked out by the mutation. Heterozygotes demonstrate no overt phenotype, but homozygotes die at birth.

Development

A cassette composed of 3XStop (TGAttaattaa)-IRES-CreERT2-s.int (synthetic intron)-WPRE-pA-FRT-PGK-neomycin-pA-FRT was introduced to exon 2 (replacing the first ATG shared by all the transcripts) via homologous recombination in 129S6/SvEvTac-derived embryonic stem (ES) cells. CreERT2 represents Cre recombinase fused to a G400V/M543A/L544A triple mutation of the human estrogen receptor ligand binding domain. Resultant mice were crossed with Actb-FlpE (Stock No. 003800) mice to excise the neomycin cassette. This strain was maintained on a mixed genetic background with contributions from 129S6/SvEvTac, C57BL/6J, and CD1 strains by the donating laboratory.

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).
