Description

Tnfaip8^flx/flx mice carry a CRISPR/Cas9 generated mutation that results in loxP sites flanking the entire coding region contained in exon 2 of the Tnfaip8 gene. These mice may be useful when studying the role of TNFAIP8 in intestinal epithelial cells and the onset of inflammatory bowel diseases such as colitis.

Detailed description

Tnfaip8^flx/flx mice carry a CRISPR/Cas9 generated mutation that results in loxP sites flanking the entire coding region contained in exon 2 of the tumor necrosis factor, alpha-induced protein 8 (Tnfaip8) gene. Tnfaip8 encodes a ubiquitously expressed protein that acts as a negative mediator of apoptosis and may play a role in tumor progression, immune homeostasis, and inflammation regulation. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 removed in the cre-expressing tissues of the offspring.

When these Tnfaip8^flx/flx mice are bred to villin::creERT2 mice (Stock No. 020282), with tamoxifen-inducible Cre recombinase expression in the gut, resulting TKO mice lack TNFAIP8 expression specifically in the intestinal epithelial cells depending on timing of tamoxifen administration. Colitis can be induced in TKO mice by supplementing dextran sodium sulfate (DSS) into their water.

Development

Plasmids encoding a guide RNA were designed to insert loxP sites flanking exon 2 of the tumor necrosis factor, alpha-induced protein 8 (Tnfaip8) gene. sgRNA and the cas9 nuclease were introduced into the cytoplasm of C57BL/6 zygotes with well recognized pronuclei. Injected embryos were transferred to pseudopregnant females. Resulting Tnfaip8^flx mice were identified by sequencing and PCR and further backcrossed to C57BL/6J mice (Stock No. 000664) for at least 5 generation to remove any off-target sites. By the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
