Neurog2^{Flag-mCherryKI} mice have a FLAG-tag and an IRES-mCherry red fluorescent protein inserted into the N-terminal STOP codon of the neurogenin 2 (Neurog2) gene. These mice are useful for tracing neural progenitor cells (NPCs).

Detailed description

Neurog2^{Flag-mCherryKI} mice have a FLAG-tag and an IRES-mCherry red fluorescent protein inserted into the N-terminal STOP codon of the neurogenin 2 (Neurog2) gene. Neurog2 encodes a proneural transcription factor that plays a role in neuronal commitment and differentiation, specifically in neural progenitor cells (NPCs), and in the differentiation and survival of midbrain dopaminergic neurons. In this strain, insertion of the Flag-IRESmCherry sequence into the STOP codon does not affect endogenous Neurog2 expression. mCherry fluorescence is detectable by FACS analysis and immunohistochemistry. These mice are useful for tracing neural progenitor cells (NPCs).

Heterozygous mice are viable and fertile. The donating investigator did not attempt to make a homozygous colony.

Development

The Neurog2^{Flag-mCherryKI} targeting vector was designed by Dr. Carol Schuurmans while at Sunnybrook Research Institute (Toronto, ON) to insert a FLAG-tag and an internal ribosome entry site (IRES) followed by an mCherry red fluorescent protein sequence and a loxP-flanked neo cassette at the STOP codon of the neurogenin 2 (Neurog2) gene. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCrl)F1-derived G4 embryonic stem (ES) cells. Correctly targeted ES cells were aggregated morulae to produce chimeras, which were bred with CD1 mice. Resulting offspring were further maintained on an outbred background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).
