Description

The pL2-IL-1B transgenic mice express a constitutively secreted form of mature human IL-1β (interleukin-1β) in the esophagus and forestomach epithelium specifically.

Detailed description

The pL2-IL-1B mice express a constitutively secreted form of mature human IL-1β (interleukin-1β) in the esophagus and stomach epithelium specifically. The donating investigator has shown that the pL2-IL-1B mice exhibited thickened esophagus and squamous forestomach with acute and chronic inflammatory infiltrates in 6-month or older animals. At 6-month-old, pL2-IL-1B mice show moderate inflammation and profound epithelial hyperplasia, and at 12-15 months of age developed severe columnar metaplasia at the squamocolumnar junction (SCJ) of the esophagus/forestomach. This line will be useful for studies in esophagitis, Barrett’s esophagus (BE), and esophageal adenocarcinoma (EAC).

Development

The pL2-IL-1B transgene consisted of a constitutively secreted form (i.e. not requiring caspase-1 cleavage) of mature human IL-1β (IL1B) cDNA fused with the signal sequence derived from the structurally related human IL1-RA (IL1RN). These sequences were subcloned along with the hGH/polyadenylation signal sequence, downstream of the EBV promoter, into the pB-KS+ vector, linearized, and the final ~550bp transgene was injected into C57BL/6 x SJL F2 hybrid zygotes to generate two founder lines. This is the high-expression line 1 with an unknown insertion site and copy numbers. All transgenic mice were backcrossed to C57BL/6J mice (Stock No. 000664) over six generations to remove other genes before being sent to The Jackson Laboratory to cryopreserve their sperm (see SNP note below). To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.



In 2025, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Four of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.

Molecular note: The Epstein-Barr virus ED-L2 promoter drives the expression of a modified human IL1B cDNA. The modified human IL1B cDNA consists of the IL1RN signal sequence and a cDNA encoding for the 17 kDa mature human IL1B. Two lines were generated one with high expression and one with low expression. This is the high expression line 1.
