Dclk1 CreERT BAC transgenic mice express the gastrointestinal epithelium-specific, tamoxifen (Tx)-inducible Cre recombinase from the mouse Dclk1 (doublecortin-like kinase 1) promoter in adult gastrointestinal tuft cells.

Detailed description

Dclk1-CreERT BAC mice express the tamoxifen (Tx)-inducible Cre recombinase from the mouse Dclk1 (doublecortin-like kinase 1) promoter in adult gastrointestinal tuft cells, specifically in pancreatic acinar progenitor, duct, and islet cells. The donating investigator has shown that the subpopulation of Dclk1-positive tuft cells is increased in inflammation-induced carcinogenesis, is long-lived, and contributes to recovery following intestinal and colonic injury showing rare stem cell abilities. This line will be useful for studies on gastrointestinal tuft cell lineage and functions, as well as colorectal tumorigenesis.

Development

Using recombineering methods, the cre/ERT2-FrtNeoFrt cassette was inserted upstream of the ATG codon in exon 2 of the Dclk1 gene in the mouse BAC clone RP23-283D6 containing approximately 50-kb 5' sequences of the Dclk1 gene-coding region. From positive clones, the Frt-flanked neo cassette was excised using arabinose before insertion of the transgene. The 199114bp modified BAC construct was injected into fertilized CBA x C57BL/6J oocytes. Four founders are positive for the transgene. Lines 2 and 4 are kept and maintained. Line 2 transgene is reported to be expressed more in the pancreas/esophageal tissues, while line 4 expresses more in the colon. This strain is line 2 with an unknown transgene copy number or genomic insertion site, which was crossed with wild-type siblings for 12 generations through selective breeding, using breeders with high Cre activity. Mice have been heavily crossed with other mutant strains but were backcrossed to C57BL/6J mice (Stock No. 000664) to remove the other genes before being sent to The Jackson Laboratory to cryopreserve the sperm. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
