TNF receptor-associated factors (TRAFs) are recruited to many TNF-receptor (TNF-R) superfamily members and are important modulators of the proximal signaling events that occur at the time of T cell receptor engagement and activation. TRAF5 (TNF receptor-associated factor 5) has been shown to be a positive regulator of a number of these receptors that are involved in T- and B-lymphocyte activation, including those mediating T cell costimulation. TRAF5- mice carry a targeted knock-out of the mouse Traf5 gene in which exon 2, encoding the C-terminus of the RING finger domain, was replaced with a neomycin cassette. Western blot analysis demonstrates an absence of intact or truncated protein in the tissue of homozygotes. TRAF5-deficient mice are born at the expected Mendelian ratios. The mice are healthy and show no obvious abnormalities through 24 weeks of age.
