Description

Fgfr1^Myc knock-in mice express fibroblast growth factor receptor 1 (FGFR1) protein with an in-frame 2xMyc epitope tag at the 3' end. This strain is useful in labeling FGFR1:FGFR2 heterodimers in a range of studies including development, cell proliferation, differentiation, and adhesion.

Detailed description

Fibroblast Growth Factor (FGF) signaling plays an integral role in development, driving numerous cellular processes including proliferation, differentiation, and cellular adhesion.

These Fgfr1^Mycknock-in mice express fibroblast growth factor receptor 1 (FGFR1) protein with an in-frame 2xMyc epitope tag at the 3' end. Myc-positive staining has been observed in embryonic (E14.5) facial mesynchymal tissues, such as the Meckel’s cartilage. This strain, in conjunction with Fgfr2^Flag mice (Stock No. 039110), is useful in detecting the presence of FGFR1:FGFR2 heterodimers in a range of research areas including immunohistochemistry, biochemical studies, and proteomics.

Development

A linearized targeting vector, used to introduce an in-frame 2xMyc epitope tag to the 3' end of the mouse Frfr1 gene, was electroporated into 129S4/SvJaeSor-derived AK7 embryonic stem (ES) cells. Resultant chimeric mice were bred to Meox2-Cre deleter mice on a 129S4/SvJaeSor genetic background to remove a loxP-flanked neomycin selection cassette that had been introduced to exon 8. The Meox2-Cre allele was subsequently crossed out. Mice were maintained on a 129S4 background by the donating laboratory.

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve 129S4/SvJaeSorJ (Stock No. 033000) or 129S4/SvJaeJ (Stock No. 009104).
