Detailed description

This strain carries a null allele for Trhr, which is valuable in characterizing thyroid function. Mice homozygous for this null allele are smaller than normal at wean age. The loxP-flanked conditional-ready allele that gave rise to this null allele is also available in Stock No. 036337 and on a 129S congenic background in Stock No. 036336.

Development

The Trhr conditional knockout construct was inserted into 129S1/SvlmJ ES cells (stock#002448) which were inserted into C57BL/6J embryos. This construct inserted a loxP site upstream of exon2 and an FRT-flanked neomycin selection cassette followed by a loxP site downstream of exon 2. Chimeric mice were mated to C57BL/6J then further backcrossed to C57BL/6J for 7 more generations. An N8 congenic male was mated to B6.C-Tg(CMV-cre)1Cgn/J (stock#006054) to create a null allele. The resulting mice that were hemizygous for the null allele were intercrossed and this strain was further maintained by mating homozygous null males to hemizygous females. Sperm was cryopreserved from heterozygous males at generation is N9F25.
