Description

Tacr1^CreERT2 knock-in/knock-out mice express tamoxifen-inducible Cre recombinase from the mouse tachykinin receptor 1 (Tacr1) promoter in Tacr1-expressing cells of the spinal cord and brain, including spinal projection neurons of the anterolateral pathway. This strain may be useful in studies of nociception (pain and affective touch).

Detailed description

Tacr1⁺ (tachykinin receptor 1) and Gpr83⁺ (G protein-coupled receptor 83) spinoparabrachial (SPB) neurons form parallel ascending circuit motifs that cooperate to convey nociceptive (pain and affective touch) signals to the brain. Strong activation of these neurons generates spatiotemporally distinct patterns of escape locomotion, autonomic (pupillary) reactions and place aversion.

These Tacr1^CreERT2 knock-in/knock-out mice express tamoxifen-inducible cre recombinase from the mouse Tacr1 promoter in all examined Tacr1-expressing cells of the spinal cord and brain, including spinal projection neurons of the anterolateral pathway. Expression of the targeted Tacr1 gene is eliminated. The mice demonstrate no overt phenotype.

Development

The first coding ATG in exon 2 of the mouse Tacr1 gene was replaced with a tamoxifen-inducible Cre recombinase-estrogen receptor T2 (CreERT2) cassette and FRT-flanked PGK-neomycin cassette via homologous recombination in 129S6/SvEvTac-derived embryonic stem (ES) cells. Resultant mice were crossed with a germline FlpE strain (Stock No. 003800) to excise the neomycin cassette. This strain was maintained on a mixed genetic background incorporating 129S6/SvEvTac, C57BL/6J, SJL, and Crl:CD1 genetic contributions by the donating laboratory.

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).
