Description

mTFP1-cRel reporter mice have the Rel gene driving expression of a monomeric turquoise fluorescent protein (mTFP1). These mice may be a useful tool for visualizing NFΚB activity in B-cell development and differentiation.

Detailed description

The Rel (reticuloendotheliosis oncogene), also known as the proto-oncogene c-Rel, encodes for a member of the Rel/NF-κB family of transcription factors which are involved in cell growth and survival, stress responses and inflammation. These mTFP1-cRel reporter mice have a monomeric turquoise fluorescent protein sequence (mTFP1), followed by a short flexible linker sequence, inserted into exon 2 of the Rel gene.

Homozygous mTFP1-cRel mice are viable and fertile with no overt phenotype; however, the donating laboratory does indicate that homozygous mice do display slightly reduced in vitro B-cell proliferation capacity compared to wildtype controls. Turquoise fluorescence can be detected by flow cytometry in homozygous or heterozygous mTFP1-cRel cells. These mice may be useful for studying NFκB activity in B-cell development and differentiation.

Development

The mTFP1-cRel knock-in (KI) targeting vector was designed to insert a turquoise fluorescent sequence upstream of exon 2 of the reticuloendotheliosis oncogene (Rel). This construct also included a short flexible linker (G4S)₂ and a FRT-flanked neomycin cassette. The targeting construct was electroporated into C57BL/6NTac-dervied IC1 embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred with mice expressing ACTB:FLPe recombinase (Stock No. 003800), to remove the neo cassette. The resulting mTFP1-cRel mice, with neo cassette removed, were then backcrossed to C57BL/6J mice for at least 6 generations by the donating laboratory (and the ACTB:FLPe was removed). Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
