FX- mice carry a targeted knock-out allele of the mouse coagulation factor 10 (F10; FX) gene in which all of the coding exons were replaced with a neomycin cassette. These mice have been useful in studies of bleeding disorders. Homozygotes demonstrate partial embryonic lethality, with approximately one third dying around embryonic day 11.5-12.5. These embryos show signs of massive bleeding, but no histological defects in the vasculature or the yolk sac. The remaining homozygous embryos appear normal and survive to term, but the majority of neonates (90%) die within 5 days, most frequently from intraabdominal bleeding. The remaining animals die between postnatal days 5-20 with intraabdominal, subcutaneous, or intracranial bleeding or a combination thereof. Heterozygotes are viable and fertile and do not present any overt phenotype.
