Description

Plin5^loxP mice have loxP sites flanking exons 3-8 of the perilipin 5 gene. These mice may be useful for conditional deletion of PLIN5 when studying white and brown adipose tissue remodeling and systemic glucose tolerance during cold exposure.

Detailed description

Plin5^loxP mice have loxP sites flanking exons 3-8 of the perilipin 5 (Plin5) gene. Plin5 encodes a lipid droplet coat protein expressed in highly oxidative tissues, such as heart, oxidative skeletal muscle, fasted liver, and adipose tissues. PLIN5 has been localized to muscle mitochondria and adipose tissues where it is expressed in inguinal and epididymal white adipose tissue (WAT), with higher expression in brown adipose tissue (BAT). The expression of Plin5 has been shown to further increase in BAT during cold exposure and PLIN5 has also been shown to influence systemic metabolism.

Homozygous mice are viable and fertile. When bred to Ucp1-cre transgenic mice (Stock No. 024670), resulting BKOPLIN5 offspring, lacking PLIN5 in BAT, display mostly preserved glucose tolerance and cold tolerance in compared to control mice, likely due to compensation that occurs in inguinal WAT. Also, during cold exposure, mitochondrial area is increased and mitochondrial length decreased in the BAT of BKOPLIN5 mice.

Development

The Plin5^loxP targeting vector was designed to insert loxP sites flanking exons 3-8 of the perilipin 5 (Plin5) gene, and a frt-flanked neomycin resistance (neo) cassette was inserted downstream of the floxed cassette. The construct was electroporated into C57BL/6J-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the chimeric mice were bred to C57BL/6J mice (Stock No. 000664). Subsequently, mice were bred to ROSA26::FLPe mice (Stock No. 009086) to remove the neo cassette. Resulting offspring were bred to C57BL/6J mice for at least 5 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
