Description

Rbm5 floxed mice possess loxP sites flanking exon 3 of the RNA binding motif protein 5 (Rbm5) gene. These mice may be useful for studying the role of Rbm5 in cell growth and apoptosis in multiple cancers and neurodegenerative diseases.

Note that a 48 SNP (single nucleotide polymorphism) panel analysis was performed on the rederived living colony at The Jackson Laboratory Repository. All SNP markers were consistent with C57BL/6J allele-type with the exception of Thy1^a and one marker on Chromosome 19 was segregating for a non-C57BL/6J marker (e.g., C57BL/6N or other inbred). These data suggest the mice sent to The Jackson Laboratory Repository could be on a mixed C57BL/6N;C57BL/6J genetic background or retain some genetic background contribution from the Thy1^a strain it was crossed to.

Detailed description

Rbm5 encodes a nuclear RNA binding protein that plays a role in the induction of cell cycle arrest and apoptosis through pre-mRNA splicing of multiple target genes and by regulating the activity of the tumor suppressor protein p53. Animals that do not express RBM5 globally are perinatal lethal; therefore, the donating laboratory has developed a conditional mutant with loxP sites flanking exon 3 of the Rbm5 gene.

Mice homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues. The donating laboratory bred these Rbm5 floxed mice to B6.Cg-Tg(Nes-cre)1Kln/J mice (Stock No. 003771) to generate a brain-targeted RBM5 knockout (KO) mouse. The RBM5 KO mice demonstrated complete loss of RBM5 protein in hippocampal tissue as determined by western blot analysis. These Rbm5 floxed mice may be useful for generating tissue-specific Rbm5 knock-out mice when studying multiple cancers and neurodegenerative diseases.

In 2023, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. All SNP markers were consistent with C57BL/6J allele-type with the exception of Thy1^a and one marker on Chromosome 19 was segregating for a non-C57BL/6J marker (e.g., C57BL/6N or other inbred). These data suggest the mice sent to The Jackson Laboratory Repository could be on a mixed C57BL/6N;C57BL/6J genetic background or retain some genetic background contribution from the Thy1^a strain it was crossed to [Gt(ROSA)26Sor^{tm2.1(UBC-flpe)Ozg} (MGI:6297051)].

Development

The Rbm5 floxed allele was generated by goGermline technology (Ozgene; Bentley DC, WA, Australia) using a targeting vector composed of an initial loxP site immediately upstream of exon 3, of the RNA binding motif protein 5 (Rbm5) gene, followed by an FRT-flanked neomycin cassette, and a second loxP site. The construct was electroporated into B6.Cg-Thy1^a-derived Bruce4 embryonic stem (ES) cells. Targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred with C57BL/6J to establish germline transmission. Progeny were crossed to Gt(ROSA)26Sor^{tm2.1(UBC-flpe)Ozg} mice (MGI:6297051) to excise the neo cassette. The Flp-expressing transgene was removed through selective breeding. The resulting heterozygous Rbm5 floxed mice, with neo cassette removed and floxed exon 3 intact, were intercrossed and maintained by the donating laboratory as homozygous for the Thy1^a allele and homozygous for the Rbm5 floxed allele prior to sending males to The Jackson Laboratory in 2016 for cryopreservation (see SNP note below). To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664). The resulting mice, heterozygous for both alleles, were bred together to be homozygous for both alleles.

The Thy1 locus determines a surface antigen present on cells of the thymus, a number of mouse leukemias, brain, and in lesser amounts on lymph node and spleen cells. The allele Thy1^a determines an antigenic specificity, Thy-1.1, found in the AKR and RF strains; the allele Thy1^b determines an antigenic specificity, Thy-1.2, found in the C3HeB/Fe and many other strains (J:5243, J:5012, J:4469). The Thy1 antigen is probably present on all T lymphocytes and absent from all B lymphocytes, and it thus serves as a valuable T-cell marker (J:5243). It is very widely used in experiments designed to determine the distribution and function of T-cells. Thy1 specifies a T-cell surface glycoprotein, T25, with a molecular weight of 25 kDa (J:5707). The protein appears to be anchored in the cell membrane by a lipid that is either phosphotidylinositol or closely related to it (J:12016). Thy1 may function in the cell membrane as a signal transduction molecule (J:8333).
