Description

Foxf1-tdTomato transgenic mice express a tdTomato reporter directed by mouse Foxf1 (forkhead box F1) promoter elements. tdTomato recapitulates endogenous expression of Foxf1. This strain may be used to visualize lung development.

Detailed description

Foxf1-tdTomato transgenic mice express the tdTomato fluorescent protein under the control of the mouse Foxf1 (forkhead box F1) promoter elements. Foxf1 is a transcription factor expressed in the lateral mesoderm during embryogenesis with continued expression in adult lungs. FOXF1 is essential for branching morphogenesis and lung development. The tdTomato reporter recapitulates endogenous expression of Foxf1. Mice carrying the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.

Development

The Foxf1-tdTomato transgenic construct contains the mouse Foxf1 gene (bacterial artificial chromosome clone, RP23-55G4) modified by BAC recombineering methods to insert the tdTomato coding sequence, a polyA sequence, and an FRT-flanked PGK neo:gb2:kanamycin into exon 1 of the Foxf1 gene. The final engineered BAC used for transgenic mouse production contained approximately 116,000 base pairs of DNA upstream from the tdTomato start site. The construct is followed by a chloramphenicol selection marker, which may have been retained in the final allele. The donating investigator indicates that BAC clone does not include any other known genes. Transient flp infection removed the neo and kanamycin cassette. The modified BAC was microinjected into the pronuclei of fertilized C57BL/6J oocytes. Founder line 912 was bred to C57BL/6J. The strain has been crossed to C57BL/6J for 10 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
