Description

In the absence of Cre recombination, these sPRR-Myc-tag transgenic mice express a green fluorescent protein (GFP) variant under the control of the CAG promoter. Upon Cre recombination, the GFP cassette, flanked by loxP sites, is removed and myc-tagged human sPRR (the soluble form of prorenin receptor encoded by ATP6AP2) is expressed under the control of the CAG promoter in cells where the Cre promoter is active. This strain is useful in studies of hypertension and furthering the characterization of sPRR’s functional roll in multiple tissues.

Detailed description

Prorenin receptor (PRR), encoded by the ATPase, H+ transporting, lysosomal accessory protein 2 (ATP6AP2) gene, is a newly discovered component of the renin angiotensin system (RAS) involved in the generation of angiotensin II. Independently, PRR also activates the mitogen activated protein kinase (p38 MAPK) and the ERK1/2 pathways. PRR can be cleaved to produce a carboxy terminal fragment (tPRR) and a soluble form (sPRR) that is released into the circulation and urine. A compelling body of literature has shown that sPRR plays a functional role in several tissues, among them the kidneys, liver, and adipose.

In the absence of Cre recombination, these sPRR-Myc-tag transgenic mice express a green fluorescent protein (GFP) variant under the control of the CAG promoter. Pups demonstrate fluorescence under UV (dark) light. Upon Cre recombination, the GFP cassette, flanked by loxP sites, is removed and myc-tagged human sPRR is expressed under the control of the CAG promoter in cells where the Cre promoter is active.

Crosses of female sPRR-Myc-tag with male Hoxb7/Cre mice (Stock No. 004692) produce progeny expressing renal collecting duct-derived human sPRR (K-HsPRR). Crosses with male adiponectin/Cre mice (Stock No. 010803) generate pups expressing adipose-derived human sPRR (Adi-HsPRR), and crosses with male mice expressing albumin/Cre (Stock No. 018961) generate animals expressing liver-derived human sPRR (Liv-HsPRR).

Development

Myc epitope-tagged human ATP6AP2 cDNA (encoding the soluble form of prorenin receptor, sPRR) was introduced to an unknown CAG-GFP vector to produce a CAG-loxP-GFP variant-loxP-HsPRR-cMyc transgenic construct. The GFP sequence does not appear to correspond to that of a traditional green fluorescent protein. The construct was introduced to C57BL/6J embryos and the resultant mice were maintained on a C57BL/6J genetic background by the donating laboratory. This is founder line 1. The integration site and copy number are unknown (September, 2023).

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).
