R26^LSL-Mfsd2a mice enable conditional, Cre recombinase-dependent overexpression of mouse MFSD2 lysolipid transporter A, lysophospholipid (Mfsd2a) upon excision of a loxP-flanked Stop cassette. This strain has been useful in studies of neurovascular coupling.

Detailed description

MFSD2 lysolipid transporter A, lysophospholipid (MFSD2A) is a sodium-dependent lysophosphatidylcholine symporter essential for docosahexaenoic acid uptake into the brain and blood-brain barrier function.

R26^LSL-Mfsd2a mice enable conditional, Cre recombinase-dependent overexpression of mouse Mfsd2a cDNA upon excision of a loxP-flanked Stop cassette. Prior to Cre recombinase expression, the mice have no overt phenotype.

Development

A CAG-loxP-3xSV40PA Stop-loxP-mouse Mfsd2a cDNA-WPRE-bGH polyA-attB-PGK-neomycin-attP cassette was introduced to the Gt(ROSA)26Sor locus via homologous recombination in 129S6 x C57BL/6J F1-derived embryonic stem (ES) cells. Resultant mice were crossed with ROSA26:φC31 recombinase mice (Stock No. 007743) to excise the neomycin cassette before being backcrossed to C57BL/6J for 13 generations by the donating laboratory (see SNP note below).

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).

In 2023, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on males sent to The Jackson Laboratory Repository. Three of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
