Detailed description

Myt1L^F floxed mutant mice possess loxP sites flanking sequence containing a fusion of exons 12 and 13 of the myelin transcription factor 1-like (Myt1l) gene. Myt1L encodes a transcription factor expressed in neuronal tissues that plays a key role in neuronal differentiation by repressing expression of non-neuronal genes during neuron differentiation. MYT1L loss of function mutations have been associated with intellectual disability and autism spectrum disorder (ASD), while MYT1L duplication has been observed in patients with schizophrenia. Mice that are homozygous for the conditional allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 12 and 13 (aa 491-566) deleted in the cre-expressing tissues.

Development

The Myt1L^F targeting vector contains a loxP site upstream of exon 12 of the myelin transcription factor 1-like (Myt1l) gene. A frt-flanked neomycin resistance (neo) cassette, followed by a second loxP site, was inserted downstream of exon 13. Exons 12 and 13 (aa 491-566) were fused together. The construct was electroporated into 129S6/SvEvTac-derived TL1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to Flpe mice to excise the neo cassette. Myt1L^F mice that retained the loxP site-flanked exons 12/13 were bred to remove the Flpe recombinase allele. Heterozygotes were crossed to generate homozygotes and resulting mice were backcrossed to outbred CD-1 mice by the donating laboratory. This mouse line contained St18^tm1.1Ggu and Myt1^tm1Ggu alleles upon arrival at The Jackson Laboratory. Sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664) and the St18^tm1.1Ggu and Myt1^tm1Ggu alleles were bred out of the colony. Mice containing the St18^tm1.1Ggu floxed allele are available as Stock No. 038502.
