Description

Zfp36l2^fl mice possess loxP sites flanking exon 2 of the Zfp36l2 (zinc finger protein 36, C3H type-like 2) gene. These floxed mice are useful for tissue specific deletion of Zfp36l2 when studying oocyte maturation.

Detailed description

Zfp36l2 (zinc finger protein 36, C3H type-like 2), like the other ZFP36 family members, encodes an mRNA-destabilizing protein that binds to AU-rich elements (AREs) in the 3' UTR of mRNAs and functions to promote the decay of targeted mRNAs. Complete KO of ZPF36L2 leads to lethal hematopoietic failure about 2 weeks after birth. Using conditional KO mice, ZPF36L2 has been shown to be involved in hematopoiesis, oocyte maturation, adipogenesis, erythroid cell differentiation, and, together with ZPF36L1, regulates thymocyte development and B cell differentiation. Another recent paper demonstrated tissue specific KO of this gene in myeloid cells

Zfp36l2^fl mice possess loxP sites flanking exon 2 of the Zfp36l2 gene. Exon 2 encodes the RNA binding domain and the majority of the protein coding region. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues resulting in a tissue specific null allele.

When combined with mice carrying a Cre recombinase that is expressed in growing oocytes (such as Tg(Zp3-cre)93Knw, Stock No. 003651), female progeny are infertile as a result of abnormal oocyte maturation and a failure to undergo normal fertilization.

Of note, the floxed Zfp36l1 allele is distributed as C57BL/6N-Zfp36l1^tm4.1Pjb/J (Stock No. 038472).

Development

The Zfp36l2^fl targeting vector was designed to insert a loxP site upstream of exon 2 and an FRT-flanked neomycin cassette followed by a second loxP site was inserted downstream of exon 2 of the zinc finger protein 36, C3H type-like 2 (Zfp36l2) locus on chromosome 17. The construct was electroporated into C57BL/6N-derived embryonic stem cells (Xenogen). Transient expression of flp recombinase removed the neo cassette. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6NTac females and mice were subsequently backcrossed for one year to C57BL/6NTac mice. The strain was maintained by homozygous matings. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Allele

Symbol: Zfp36l2^tm3.1Pjb
Name: targeted mutation 3.1, Perry J Blackshear
MGI accession ID: MGI:7312040
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); No functional change
Synonyms: Zfp36l2^fl
Marker symbol: Zfp36l2
Marker name: zinc finger protein 36, C3H type-like 2
Marker synonyms: Brf2; ERF2; ERF-2; OOMD13; OZEMA13; RNF162C; TIS11D
Chromosome: 17
Strain of origin: C57BL/6
Molecular note: A loxP site was inserted upstream of exon 2 and an frt-flanked neomycin selection cassette and second loxP site were inserted downstream of exon 2. The neomycin selection cassette was removed via flp-mediated recombination, leaving exon 2 floxed.
