Description

Zfp36l1^flox mice possess loxP sites flanking exon 2 of the Zfp36l1 (zinc finger protein 36, C3H type-like 1) gene. These floxed mice are useful for tissue specific deletion of Zfp36l1 when studying metabolism.

Detailed description

Zfp36l1 (zinc finger protein 36, C3H type-like 1), like the other ZFP36 family members, encodes an mRNA-destabilizing protein that binds to AU-rich elements (AREs) in the 3' UTR of mRNAs, and functions to promote the decay of targeted mRNAs. Complete KO of ZFP36L1 leads to embryonic lethality at about E10 from failure of chorio-allantoic fusion. Other studies have shown that ZFP36L1 is involved in cholesterol metabolism, thermogenesis, and liver bile acid metabolism.

These Zfp36l1^flox mice possess loxP sites flanking exon 2 of the Zfp36l1 gene. Exon 2 encodes the tandem zinc finger domain responsible for RNA binding activity. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues resulting in a tissue specific null allele.

When combined with mice carrying a Cre recombinase that is expressed in liver cells (such as Speer6-ps1^{Tg(Alb-cre)21Mgn}, Stock No. 018961), progeny fed a Lieber-DeCarli liquid diet containing alcohol are resistant to alcohol-induced hepatic steatosis, injury and inflammation and have an increase in the expression of Fgf21 in liver.

Of note, the floxed Zfp36l2 allele is distributed as C57BL/6N-Zfp36l2^tm3.1Pjb/J (Stock No. 038474).

Development

The Zfp36l1^flox targeting vector was designed to insert one loxP site upstream of exon 2 and a loxP-flanked neomycin cassette downstream of exon 2 of the zinc finger protein 36, C3H type-like 1 (Zfp36l1) locus on chromosome 12. The construct was electroporated into C57BL/6-derived embryonic stem cells (Xenogen). The neo cassette was removed by the electroporation of Cre recombinase into the correctly targeted ES cells. ES cells with neo removed were injected into blastocysts. Chimeric males were crossed to C57BL/6NTac females, and mice were subsequently backcrossed for one year to C57BL/6NTac mice. The strain was maintained by homozygous matings. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).
